John Chamerlain Holden married his cousin, Grace Holden, 20 December 1813, in Harrison County, West Virginia. Grace was born 10 November 1790, in New Jersey, daughter of Benjamin and Mary Jemima (Derumple) Holden. They had three children together. She died 16 May 1819, in Harrison Co., WV.

John married Prudence Kittle on 03 Nov or Dec 1819. She was born on 24 Apr 1801 to Margaret (Marteney) and Abraham Kittle, Jr. John and Prudence had 15 children together.
